The Brazilians went into half-time 4-0 ahead.
A number of changes from Sundowns saw Rushine De Reuck given his first start while goalkeeper Ricardo Goss, Thapelo Morena, Haashim Domingo and Lesedi Kapinga were also recalled.
It took the Brazilians just 10 minutes to take the lead after goalkeeper Tebatso Phoku parried a shot from Haashim Domingo into the path of Peter Shalulile who found the top-corner.
Four minutes later Mamelodi Sundowns doubled their lead when possession was turned over allowing Lebohang Maboe to run in-behind for a simple finish.
Sundowns made it 3-0 a minute later when Lesedi Kapinga scored.
The Brazilians added a fourth in the 28th minute from the penalty spot through Andile Jali after Kapinga was fouled inside the box.
Mamelodi Sundowns will now play Orlando Pirates in the quarter-finals on Thursday, 15 April.
Kick-off is at 18:00.
